Histostar(Ms + Rb) (for Human tissue)
Histostar™ is a two-step IHC staining reagent. This reagent is based on an horseradish peroxidase (HRP) labeled polymer which is conjugated with secondary antibodies. Histostar™ (Ms + Rb) is designed to allow Immunohistochemical studies using a user-supplied Mouse primary antibody and Rabbit primary antibody. This reagent is available for formalin fixed paraffin-embedded human tissue sections.
Anti-Mouse and Anti-Rabbit) is the labeled polymer prepared by combining amino acid polymers with peroxidase and goat anti-Mouse Ig and goat anti-Rabbit Ig which are reduced to Fab’. IgG fraction purified from immunized goat serum is digested to prepare F(ab’)2. Antigen-specific F(ab’)2 is affinity-purified with the antigen. Solid-phase absorption is carried out with human serum protein. Peroxidase-labeled amino acid polymer is conjugated to Fab’ obtained by reducing F(ab’)2.

Description/Background: Immunohistochemistry (IHC) is an efficient means to understand antigens distribution and localization in cells or tissue sections under the microscope. IHC is widely used in clinical diagnosis of disease and basic research purposes. IHC uses monoclonal or polyclonal antibodies to determine the tissue distribution of an antigen of interest. The site of antibody binding is visualized by a marker such as fluorescent dye, enzyme, radioactive element or colloidal gold that is directly linked to the primary antibody or to an appropriate secondary antibody. In immunoenzyme technique, frequently used streptavidin-biotin complex (SAB) methods or avidin-biotin complex (ABC) method, which need additional two steps containing both Biotinylated secondary antibody reaction and peroxidase labeled streptavidin reaction after first antibody reaction. HistostarTM is a two-step IHC staining reagent. This reagent is based on a horseradish peroxidase (HRP) labeled polymer which is conjugated with secondary antibodies. It increases the number of available HRP at the antigenic site, thus increasing their reactivity with the chromogen. HistostarTM is an extremely sensitive reagent and save time. Because HistostarTM does not contain avidin and biotin, nonspecific staining as a result of endogenous biotin is eliminated.
